BTLA

BTLA,即B和T淋巴细胞弱化因子,别名BTNL9、BTLA。它是一种主要在T细胞、B细胞和NK细胞上表达的免疫检查点分子,由人体细胞自身的基因编码。BTLA的作用机制涉及与其特异性配体HVEM(疱疹病毒入侵介质)结合,这种结合能够抑制T细胞和B细胞的活化、增殖以及细胞因子的生成。BTLA与HVEM的结合通过激活免疫受体酪氨酸的抑制基序(ITIM)和免疫球蛋白酪氨酸开关基序(ITSM),导致SHP-1和SHP-2的磷酸化和募集,从而抑制T细胞的增殖和IL-2的产生,产生免疫抑制效应 。BTLA参与了多种生理和病理过程的调节,包括肿瘤、炎症性疾病、自身免疫性疾病等。在肿瘤微环境中,BTLA的激活是PD-1抗体的潜在耐药机制之一,BTLA的表达与肿瘤的进展密切相关。在炎症性疾病中,BTLA对T细胞介导的炎症具有负调节作用。此外,在自身免疫性疾病中,BTLA的表达水平与疾病的活动性相关。因此,BTLA是免疫学研究和相关疾病治疗中的一个潜在靶点 。
