AREG

AREG(Amphiregulin),全称双向调节素,也被称为双调蛋白或表皮生长因子受体配体,是表皮生长因子家族的重要成员。AREG通过与细胞表面的EGFR蛋白结合,激活下游信号网络,参与调控细胞增殖、分化及组织修复过程。研究发现,AREG在多种恶性肿瘤中异常活跃,例如结直肠癌、非小细胞肺癌和三阴性乳腺癌,其过表达会加速肿瘤生长、转移及化疗耐药。AREG蛋白在哮喘、特应性皮炎等慢性炎症疾病中可增强免疫细胞活性,并与特发性肺纤维化的异常成纤维细胞活化密切相关。目前针对AREG相关通路的治疗药物以EGFR靶向药为主,如西妥昔单抗、帕尼单抗等已用于临床,但直接抑制AREGF的药物研发仍处于早期阶段。近年来,联合靶向AREG与免疫检查点的疗法在动物模型中显示出协同抗肿瘤效果,为克服EGFR抑制剂耐药提供了新方向。
