PTDSS1
PTDSS1(磷脂酰丝氨酸合成酶1)是细胞内磷脂代谢的关键酶,也被称为PSS1或PTDSS。该蛋白通过催化丝氨酸与CDP-二酰基甘油的结合反应,在内质网中合成磷脂酰丝氨酸(PS),后者是细胞膜的重要组分。PS不仅参与膜结构的动态平衡,还在凋亡细胞表面暴露时触发免疫清除信号,并与凝血因子活化、突触可塑性等生理过程相关。近年来研究发现,PTDSS1基因突变与遗传性小脑共济失调2型(SCA2)及Lenz-Majewski综合征相关,其表达异常可能影响神经细胞膜稳定性。在肿瘤领域,PTDSS1过表达被证实通过改变细胞膜磷脂组成促进肝癌、乳腺癌侵袭转移。针对该靶点的药物研发尚处早期,已有研究尝试利用小分子抑制剂干扰PS合成通路,或通过调控PTDSS1表达增强化疗药物敏感性。2021年《自然·化学生物学》报道了首个选择性靶向PTDSS1的化合物探针,为相关疾病治疗提供了新思路。

IHC image of CSB-PA04305A0Rb diluted at 1:150 and staining in paraffin-embedded humanpancreatic cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB. Secondary antibody only control: uses 1% BSA instead of primary antibody
