TNFSF10

TNFSF10（肿瘤坏死因子超家族成员10）又称TRAIL（肿瘤坏死因子相关凋亡诱导配体），是一种Ⅱ型跨膜蛋白，其可溶性形式通过激活死亡受体DR4和DR5触发细胞凋亡。TNFSF10在免疫监视中起关键作用，可选择性诱导癌细胞凋亡而不损伤正常组织，这一特性使其成为肿瘤治疗的热点靶点。其作用涉及线粒体途径和Caspase蛋白酶级联反应，同时受凋亡抑制蛋白c-FLIP和Bcl-2家族调控。
TRAIL在肝癌、肺癌等多种恶性肿瘤中表达异常，其信号通路失调与肿瘤耐药密切相关。近年来，针对TRAIL受体的单克隆抗体如Conatumumab、Lexatumumab已进入临床试验，但单药疗效有限，现多探索与化疗或靶向药的联合应用。另有研究揭示TRAIL在阿尔茨海默病中可能通过清除异常蛋白沉积发挥保护作用，但过度激活也可能导致神经元损伤。目前新型融合蛋白如TRAIL-Fc和三特异性抗体正在开发中，旨在提高受体结合特异性和克服耐药性瓶颈。

Immunohistochemistry of paraffin-embedded human lung tissue using CSB-PA07475A0Rb at dilution of 1:100

Immunofluorescence staining of A549 cells with CSB-PA07475A0Rb at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
