TMED4

TMED4,即跨膜p24运输蛋白4。别名HNLF、ERS25、p24a3。TMED4在多种细胞过程中发挥作用,包括内质网应激反应、热休克反应、细胞凋亡以及囊泡蛋白运输,主要在早期分泌途径中。
TMED4的作用机制涉及内质网应激(ERS)的调节,通过IRE1α-XBP1信号通路和NRF2相关的抗氧化反应,促进调节性T细胞(Treg)中活性氧(ROS)的稳态,维持Treg细胞Foxp3的表达及抑制功能。TMED4对于维持Treg细胞的稳定性和功能至关重要,其缺失会导致Treg细胞中Foxp3稳定性降低,影响Treg细胞的抑制功能,进而可能导致T细胞过度激活和炎症反应加剧。TMED4还可能通过ER相关的降解(ERAD)系统抑制IRE1α的蛋白酶体降解。TMED4的功能异常与多种疾病相关,包括神经细胞脂褐素沉积症8型。
