TFR2

TFR2,即转铁蛋白受体2,别名HFE3、TFRC2。它是一种单跨膜蛋白,属于转铁蛋白受体样家族的成员,主要在肝脏和肠道细胞中表达。TFR2的作用机制涉及调节细胞对结合转铁蛋白的铁的摄取,可能参与铁代谢、肝细胞功能和红细胞分化。TFR2在铁稳态调节中发挥重要作用,其表达水平不受细胞内铁状况的调节,且在肝细胞、红系前体细胞、K562细胞和HepG2细胞中高水平表达。TFR2的突变可导致遗传性血色病III型的发生,这表明它在铁代谢中的重要性。TFR2还可能在红系前体细胞的分化发育中发挥作用,是红系前体细胞的一种分子标志。

IHC image of CSB-PA14729A0Rb diluted at 1:200 and staining in paraffin-embedded human liver cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunofluorescence staining of MCF-7 cells with CSB-PA14729A0Rb at 1:66, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
