TET2
TET2,甲基胞嘧啶双加氧酶2,别名Ten-eleventranslocation2(TET-2)。它是一种α-酮戊二酸和Fe2+依赖的双加氧酶,主要来源于哺乳动物的TET蛋白家族,该家族包括TET1、TET2和TET3三个成员。TET2的作用机制是通过催化5-甲基胞嘧啶(5mC)氧化生成5-羟甲基胞嘧啶(5hmC),进而通过一系列的氧化步骤最终转化为未修饰的胞嘧啶,这一过程是DNA主动去甲基化的重要途径。TET2在细胞命运决定、分化、肿瘤发生发展以及免疫调控中发挥重要作用。TET2的异常表达或突变与多种疾病相关,包括髓系肿瘤和免疫相关疾病。在肿瘤抑制方面,TET2通过调控基因表达和DNA甲基化状态,影响细胞增殖和分化。在免疫调控中,TET2能够影响炎症因子的分泌,参与炎症反应的调控。
