SIRT6
SIRT6全称为沉默调节蛋白6,是sirtuin家族成员之一,因其依赖辅酶NAD+发挥去乙酰化酶活性,又被称作NAD依赖性组蛋白去乙酰化酶。SIRT6在基因组稳定性维持和代谢调控中发挥核心作用,通过修复DNA损伤、调控糖脂代谢相关基因表达延缓细胞衰老进程。研究发现SIRT6缺失会引发早衰综合征,其表达异常与肝癌、结肠癌等恶性肿瘤转移密切相关,同时在Ⅱ型糖尿病、动脉粥样硬化等代谢性疾病中呈现显著下调趋势。近年研究发现该蛋白可抑制NF-κB通路减轻慢性炎症,并通过激活AMPK通路改善胰岛素抵抗。当前针对SIRT6的药物开发聚焦于小分子激活剂,如化合物MDL-800已在动物模型中证实可增强DNA修复能力并延长寿命,而部分天然产物如白藜芦醇衍生物也显示出调控潜力。随着PROTAC等新技术的应用,靶向SIRT6的双功能分子正成为抗衰老和癌症治疗领域的研究热点。
