ROS1

ROS1,即c-ros肉瘤致癌因子-受体酪氨酸激酶,别名CD74-ROS1、EZR-ROS1、SLC34A2-ROS1等,这些别名通常表示ROS1基因与其他基因发生融合的形式。ROS1基因定位于人类6号染色体长臂,属于酪氨酸激酶胰岛素受体基因,由胞内激酶区、跨膜区及胞外区三部分组成,编码具有酪氨酸激酶活性的跨膜蛋白 。ROS1的作用机制涉及其作为细胞表面受体的功能,当接收到外部特定信息后活化,并传递信号到细胞内部,指导细胞有序地生长和增殖。若ROS1基因发生致癌变异,会激活其下游通路的信号,造成细胞过度地生长及增殖,驱动肿瘤的发生。ROS1主要参与的信号通路包括RAS-RAF-MEK-ERK(MAPK)、PI3K-AKT-mTOR等 。在生物学意义上,ROS1基因融合在多种肿瘤中被认为是致癌因素,特别是在非小细胞肺癌中,ROS1融合基因型肺癌的发病率较低,但具有独特的临床病理特征。ROS1融合基因的发现为靶向治疗提供了新的治疗靶点,已有多种针对ROS1的小分子抑制剂在临床试验中显示出疗效,如克唑替尼等 。

Immunofluorescence staining of Hela cells with CSB-PA020072LA01HU at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
