RNF43
RNF43(环指蛋白43)是一种位于细胞膜表面的E3泛素连接酶,在调控Wnt信号通路中发挥关键作用。RNF43通过标记细胞膜上的Frizzled受体进行泛素化降解,从而抑制Wnt信号过度激活,维持细胞增殖与分化的平衡。RNF43基因突变常见于结直肠癌、子宫内膜癌及胰腺导管腺癌,尤其在微卫星不稳定型肿瘤中高频出现,其功能缺失会导致Wnt/β-catenin通路持续活化,促进肿瘤发生。近年来针对RNF43突变肿瘤的治疗策略取得进展,如Wnt抑制剂LGK974(PRI-724)已在临床试验中显示对携带RNF43失活突变的胰腺癌患者具有潜在疗效。基于CRISPR技术修复RNF43功能或联合PARP抑制剂的新型疗法也在探索中,为相关恶性肿瘤提供了精准治疗方向。

IHC image of CSB-PA019892LA01HU diluted at 1:600 and staining in paraffin-embedded human lung cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
