PTPN22
PTPN22，即蛋白酪氨酸磷酸酶非受体22型。别名PEST-Domain Phosphatase、PTPN8、PEP。PTPN22基因位于1号染色体上，表达产物为Protein Tyrosine Phosphatase Non-Receptor Type 22。PTPN22是一种淋巴细胞特异性的细胞内磷酸酶，与分子适配器蛋白CBL相关，可能参与调节T细胞受体信号通路中的CBL功能。该基因的突变与多种自身免疫性疾病有关，如1型糖尿病、类风湿关节炎、系统性红斑狼疮和Graves病。PTPN22在调节T细胞信号传导中起关键作用，通过去磷酸化T细胞信号通路中的关键激酶如Lck、ZAP70、Vav和EB1来抑制T细胞抗原受体（TCR）信号传导。此外，PTPN22还在血小板功能和动脉血栓形成中发挥负调控作用，PTPN22缺乏会显著缩短尾部出血时间并加速动脉血栓形成，而不会影响静脉血栓形成和凝血因子VIII和IX。
