PTAFR

血小板激活因子受体(Platelet-Activating Factor Receptor,PAFR)是一种G蛋白偶联受体,广泛分布于血小板、中性粒细胞及血管内皮细胞表面。该受体因介导炎症级联反应而闻名,当与配体PAF结合后,可触发细胞内钙离子释放并激活磷脂酶C,进而促进血管通透性增高、白细胞趋化及血小板活化。其异常活化与急性呼吸窘迫综合征、过敏性休克、动脉粥样硬化等病理过程密切相关,在脓毒症中常作为促炎介质加剧全身炎症反应。
目前全球已开发多款PAFR拮抗剂,如昔帕芬旦(TCV-309)和来帕泛(Lexipafant)曾进入败血症临床试验,但因疗效未达预期而终止。近年研究转向该受体在肿瘤微环境中的作用,发现其通过调控NF-κB通路促进乳腺癌转移,相关小分子抑制剂正在临床前评估中。某些抗组胺药物被发现具有非特异性PAFR阻断效应,这为哮喘等过敏性疾病治疗提供了新的联合用药思路。
