NRG1

NRG1,即神经调节蛋白1,别名HRG、NDF、GGF。这是一种由人类NRG1基因编码的蛋白质,属于表皮生长因子(EGF)家族。NRG1主要是通过其异构体与ErbB家族受体结合,激活多重下游信号通路,如PI3K/Akt、MAPK/ERK等,从而促进细胞增殖、分化,抑制细胞凋亡,调节能量利用,并增强血管生成。NRG1的表达和功能异常与多种疾病的发生发展相关,包括肿瘤、心血管疾病、神经精神疾病等。在肿瘤领域,NRG1基因融合是一种新兴的、潜在的致癌驱动因素,与多种肿瘤的发生发展相关,如非小细胞肺癌、肾癌、甲状腺癌等。在心血管疾病中,NRG1/ErbB信号通路在心脏的发育、心肌增殖分化及心脏传导系统的形成过程中起到了重要作用,并在心血管急慢性疾病中对心肌细胞具有保护功能。因此,NRG1被认为是这些疾病潜在的治疗靶点。
