MAG

髓鞘相关糖蛋白(Myelin-associated glycoprotein,MAG)是中枢神经系统髓鞘的重要组成部分,也被称为Siglec-4a。它由少突胶质细胞分泌,主要分布在髓鞘最内层,通过与神经元表面神经节苷脂GD1a/GT1b结合,在维持轴突髓鞘结构稳定中起关键作用。MAG具有抑制神经再生的特性,尤其在脊髓损伤后,MAG与Nogo受体结合后激活下游信号,可阻碍受损轴突的延伸,这一特性使其成为神经修复研究的重点靶点。
MAG异常表达与多发性硬化、脊髓损伤后遗症等脱髓鞘疾病密切相关,其过度抑制作用可能加剧神经功能障碍。近年针对MAG的抗体药物开发取得进展,例如通过阻断MAG与其受体的相互作用来促进神经再生。目前相关小分子抑制剂和单抗已进入临床前评估阶段,为治疗中枢神经损伤提供了新方向。阿尔茨海默病患者脑脊液中检测到MAG水平异常升高,提示其可能参与神经退行性病变过程。

Immunofluorescence staining of MCF-7 cells with CSB-PA013322LA01HU at 1:200, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
