LILRB1

LILRB1,即白细胞免疫球蛋白样受体B1,别名ILT2(免疫球蛋白样转录物2)、LIR1(白细胞免疫受体1)、CD85j。LILRB1是一种主要在髓系细胞上表达的抑制性受体,它通过与MHC I类分子等配体结合来调节免疫细胞的活性。LILRB1的作用机制涉及到其胞内结构域中的免疫受体酪氨酸抑制基序(ITIM),当LILRB1与其配体结合时,可以招募含有磷酸酶SHP-1(Src同源区域2结构域含有的磷酸酶1)的分子,从而抑制过度的免疫反应和促进免疫耐受。LILRB1在生物学上的意义主要体现在其对免疫应答的调节作用。它在树突状细胞的分化和功能中发挥关键作用,能够影响树突状细胞分泌细胞因子的能力,如IL-10、IL-12p70和TGF-β,进而调节T细胞的激活和增殖。此外,LILRB1还在肿瘤免疫微环境中起到重要作用,其表达在肿瘤相关巨噬细胞(TAM)上被上调,通过与肿瘤细胞表面的MHC I类分子结合,可能帮助肿瘤细胞逃避免疫系统的攻击。因此,LILRB1不仅在维持免疫平衡中发挥重要作用,也是癌症免疫治疗中的潜在靶点。

IHC image of CSB-PA847747LA01HU diluted at 1:400 and staining in paraffin-embedded human tonsil tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
