KLRG1

杀伤细胞凝集素样受体G1(KLRG1)又称CLEC15A,是表达于自然杀伤细胞和T细胞表面的免疫抑制性受体。KLRG1通过识别E-钙黏蛋白等配体传递抑制信号,参与调控免疫细胞活化阈值,在维持免疫稳态中起重要作用。慢性病毒感染患者和肿瘤微环境中的T细胞常呈现KLRG1高表达,这种终末分化表型与细胞增殖能力下降密切相关,可能影响抗病毒或抗肿瘤免疫应答。在自身免疫性疾病领域,KLRG1阳性调节性T细胞亚群显示出更强的免疫抑制功能,其动态变化与多发性硬化等疾病进程存在关联。近年针对免疫检查点的药物开发中,KLRG1作为潜在靶点受到关注,已有研究团队开发出阻断性抗体,在黑色素瘤等动物模型中观察到联合治疗可增强PD-1抑制剂疗效。但相较于成熟靶点,其转化医学研究仍处于早期阶段,配体互作网络与下游信号传导的具体机制尚待深入解析。

The cells were incubated in 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
