KIF18A

KIF18A,即驱动蛋白家族成员18A。别名CD319。它在细胞内主要位于细胞质和细胞核中,由898个氨基酸残基组成,分子量约为100kDa。KIF18A的作用机制涉及其作为kinesin-8家族成员,依靠水解ATP释放的能量,以微管为轨道向正极方向运动,调控微管的动态不稳定性,发挥类似于微管解聚酶的活性。KIF18A对维持染色体稳定性和控制有丝分裂过程中的染色体排列至关重要,其功能异常可能导致细胞转化和致癌。多项研究表明,KIF18A在多种癌细胞中具有高表达,与乳腺癌、透明细胞肾癌和结肠癌患者的预后相关,参与肝细胞癌的侵袭和转移,并与细胞增殖、肿瘤分期和多发性肿瘤的预后相关。KIF18A是一种可能的癌症预后生物标志物,也是抗有丝分裂治疗的潜在靶点。

Immunofluorescence staining of A549 cells with CSB-PA847753LA01HU at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
