KIF11
KIF11(驱动蛋白家族成员11)又称Eg5或Kinesin-5,是一种微管相关马达蛋白,通过水解ATP产生机械力推动微管滑动。它在有丝分裂中驱动双极纺锤体的形成与分离,确保染色体精确分配到子细胞,若功能异常可能导致染色体非整倍性,进而引发细胞癌变或凋亡。该蛋白异常高表达常见于乳腺癌、结直肠癌等实体瘤,与肿瘤增殖、转移及不良预后相关。此外,KIF11基因突变可导致常染色体显性小头畸形、视网膜变性等罕见神经发育疾病。
针对KIF11的抗癌药物研发聚焦于抑制其ATP酶活性,代表性化合物如ispinesib、filanesib通过阻断纺锤体分离迫使癌细胞停滞在分裂中期。尽管早期临床试验在血液肿瘤中显现疗效,但实体瘤响应有限且存在神经毒性问题。近年研究转向联合用药策略,如Eg5抑制剂与紫杉醇联用可增强抗微管药物敏感性。新型变构抑制剂ARRY-520已完成II期试验,而PROTAC技术开发的降解剂正探索克服耐药性的可能。
