ITGB4

整合素β4(ITGB4)是整合素家族成员,与α6亚基结合形成α6β4复合物,在细胞黏附和信号转导中起关键作用。ITGB4也被称为CD104,主要分布于上皮细胞半桥粒结构,负责锚定细胞与基底膜间的连接。ITGB4可通过激活PI3K和Ras通路参与细胞迁移及生存调控,其异常表达与多种侵袭性肿瘤密切相关,如三阴性乳腺癌、胰腺癌中常出现ITGB4过表达,促进肿瘤转移和耐药性产生。ITGB4基因突变会导致交界型大疱性表皮松解症,患者皮肤出现严重水疱病变。近年来针对该蛋白的药物开发聚焦于单克隆抗体和肽类抑制剂,已有抗ITGB4人源化抗体(如ASC-06)进入临床前研究,拟用于抑制肿瘤细胞侵袭。另有研究探索其与EGFR抑制剂联用方案,旨在突破现有靶向治疗的耐药瓶颈。

Overlay Peak curve showing A549 cells surface stained with CSB-RA011887MA1HU (red line) at 1:100. Then 10% normal goat serum was Incubated to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45 min at 4°C. The secondary antibody used was FITC-conjugated Goat Anti-Mouse IgG(H+L) at 1/200 dilution for 35 min at 4°C. Isotype control antibody (green line) was mouse IgG1 (1µg/1*106cells) used under the same conditions. Acquisition of >10, 000 events was performed.
