HYAL2
透明质酸酶2(Hyaluronidase 2,HYAL2)是人体内降解透明质酸的关键酶之一,常被称为Hyal2或PH-20样酶。该酶主要与HYAL1协同作用于溶酶体,将高分子量透明质酸切割为20-25个糖单位的片段,其活性依赖于酸性环境。HYAL2可通过调控细胞外基质重塑参与组织修复和肿瘤微环境形成,其异常表达与前列腺癌、膀胱癌等恶性肿瘤的侵袭转移密切相关,在乳腺癌中还被发现能通过激活EGFR信号通路促进耐药性形成。HYAL2在心血管疾病领域也受到关注,其介导的透明质酸降解可能影响动脉粥样硬化斑块稳定性。针对透明质酸酶家族的药物开发多聚焦于癌症治疗,例如新型小分子抑制剂SW-173T已进入临床前研究,可通过阻断HYAL2活性抑制肿瘤细胞迁移。由于透明质酸酶家族成员的功能冗余性,针对HYAL2的特异性靶向药物仍面临挑战。
