HAS1

透明质酸合成酶1(Hyaluronan Synthase 1,简称HAS1)是合成透明质酸的关键酶之一,也被称为HA synthase或细胞外基质调节因子。HAS1通过催化葡萄糖醛酸和N-乙酰葡萄糖胺的聚合,生成透明质酸分子,这种黏多糖广泛存在于皮肤、关节液和细胞间质中,具有维持组织水合度、促进创伤修复等功能。HAS1的异常表达与多种病理过程密切相关。在肿瘤微环境中,其过度活跃可能加速癌细胞转移,尤其在乳腺癌和前列腺癌中表现突出;而在类风湿性关节炎患者中,关节滑膜细胞的HAS1活性升高会导致透明质酸碎片堆积,引发炎症反应。近年来针对该靶点的药物开发集中在抑制剂领域,例如通过小分子化合物干扰酶活性,已有部分候选药物进入Ⅱ期临床试验,用于治疗纤维化疾病和特定实体瘤。此外,基于透明质酸的皮肤填充剂和关节润滑剂已在临床广泛应用,但其长效性和稳定性仍依赖对HAS1调控机制的深入研究。
