GFRA1

GFRα1(胶质细胞源性神经营养因子受体α1)是GDNF家族配体的核心受体之一,通过与RET酪氨酸激酶受体形成复合物调控神经元存活与分化。该蛋白在胚胎发育期对交感神经、肾脏及肠道神经系统的形成起关键作用,其功能异常可能引发先天性巨结肠症、多巴胺能神经元退化等疾病。近年研究发现GFRα1在甲状腺癌、神经母细胞瘤中存在异常表达,其介导的RET信号过度激活与肿瘤侵袭转移密切相关。针对这一通路开发的凡德他尼、卡博替尼等RET抑制剂已获批用于甲状腺髓样癌治疗,而基于GDNF-GFRα1系统的基因疗法在帕金森病动物模型中展现出神经保护潜力。目前重组GDNF蛋白的鞘内给药方案正在进行Ⅱ期临床试验,旨在为肌萎缩侧索硬化等神经退行性疾病提供新治疗策略。
