FPGS

FPGS,即叶酰聚谷氨酸合酶,别名Folylpoly-gamma-glutamatesynthetase, 是一种在人类细胞中表达的酶,对于叶酸稳态和增殖细胞的存活至关重要。FPGS的作用机制是通过催化ATP依赖的谷氨酸部分添加到叶酸和叶酸衍生物中,从而在细胞溶质和线粒体内建立和维持叶酰聚谷氨酸浓度。这种酶的活性对于甲氨蝶呤(MTX)类药物的细胞内代谢过程非常关键,它催化形成MTX多聚谷氨酸,这是MTX发挥抗肿瘤作用的活性形式。FPGS的异常表达或活性降低可能导致MTX耐药,在癌症治疗中,FPGS的表达和活性是影响药物敏感性的重要因素。

Overlay Peak curve showing HepG2 cells stained with CSB-RA983477A0HU (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1µg/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG(H+L) at 1:200 dilution for 35min at 4℃.Control antibody (green line) was rabbit IgG (1µg/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
