EPHA5

EPHA5蛋白全称为促红细胞生成素肝细胞受体A5,属于Eph受体酪氨酸激酶家族成员,在胚胎发育阶段主导神经轴突导向与突触形成。EPHA5通过与细胞膜表面的ephrin-A配体结合,触发双向信号传导,调控神经细胞迁移和肿瘤微环境重塑。EPHA5在阿尔茨海默病患者海马区呈现异常低表达,其介导的突触修剪功能失调可能与tau蛋白过度磷酸化存在关联。在非小细胞肺癌和胶质母细胞瘤中,EPHA5的高表达与EGFR信号通路形成交叉调控,促进肿瘤细胞侵袭转移。针对该靶点的药物开发聚焦于双特异性抗体技术,通过阻断Ephrin-A5/EPHA5相互作用抑制肿瘤血管生成,已有两款候选药物进入临床前安全性评价阶段。该受体在视网膜退行性病变中表现出神经保护作用,相关基因疗法正在探索其在黄斑变性治疗中的应用潜力。
