EPHA4
EPHA4全称为促红细胞生成素肝细胞受体A4，属于Eph受体酪氨酸激酶家族成员，常被称为EPH受体A4或酪氨酸蛋白激酶受体Tyro3。EPHA4通过与其配体ephrin相互作用，在神经系统发育中发挥关键作用，尤其参与调控轴突导向和神经元连接的形成。EPHA4异常表达与肌萎缩侧索硬化症（ALS）密切相关，其活性降低可能加速运动神经元退化进程，相关基因疗法已在动物模型中显示出延长生存期的效果。在肿瘤领域，EPHA4在结直肠癌、胶质母细胞瘤等恶性肿瘤中呈现高表达特征，通过影响肿瘤细胞侵袭转移能力促进疾病进展。针对该靶点的药物开发聚焦于小分子抑制剂和单克隆抗体，其中选择性抑制剂ALW-II-41-27在临床前研究中展现出抑制肿瘤转移的潜力，而基于CRISPR技术的基因编辑疗法正探索其在神经退行性疾病中的应用。
