EDNRA

内皮素受体A型(Endothelin Receptor Type A,简称EDNRA)是一种G蛋白偶联受体,也被称为ET-A或ETAR,主要分布于血管平滑肌细胞。EDNRA选择性结合内皮素-1(ET-1)后,通过激活磷脂酶C和钙离子信号引发血管强烈收缩,同时促进细胞增殖与纤维化,其异常激活与肺动脉高压、雷诺氏综合征密切相关。在肿瘤微环境中,EDNRA的高表达与前列腺癌、卵巢癌的侵袭转移相关,可能通过调控血管新生和基质重塑促进肿瘤进展。
目前全球已上市选择性EDNRA拮抗剂包括安贝生坦(治疗肺动脉高压)和西他生坦,但后者因肝毒性退市。2023年进入III期临床的Zibotentan联合疗法在糖尿病肾病领域展现潜力。研究发现EDNRA抑制剂联合免疫检查点药物可改善肿瘤治疗效果,但需警惕体位性低血压等副作用。该靶点的新剂型开发与联合用药策略成为近年研究热点。
