DYRK4

DYRK4(双特异性酪氨酸磷酸化调节激酶4)属于DYRK激酶家族,在细胞周期调控和应激响应中具有关键作用。该蛋白通过磷酸化下游靶点调控细胞存活与分化,尤其在癌细胞中常通过抑制凋亡通路维持肿瘤细胞增殖。DYRK4异常表达与肝癌、胰腺癌等实体瘤进展相关,其过度激活可能加速肿瘤细胞对化疗药物的耐受性。在神经系统中,DYRK4与阿尔茨海默病相关的tau蛋白异常磷酸化存在潜在关联,可能参与神经纤维缠结的形成。针对DYRK4的药物开发,部分研究机构已筛选出选择性激酶抑制剂,如INDY和化合物5-碘雷帕霉素衍生物,这些候选药物在体外实验中显示出抑制肿瘤生长的潜力。
