CTCF

CTCF,即CCCTC结合因子,别名11-zinc finger protein、CTCFL paralog。CTCF是一种多功能转录因子,含有11个高度保守的锌指结构域,能够与DNA结合并发挥功能,如介导募集cohesin、参与X染色体失活、异染色质边界的形成、V(D)J重组、RNA剪切、凋亡等。CTCF通过识别特定的DNA序列基序结合到染色质上,并作为锚点固定染色质环,是构建染色质三维结构的基础。CTCF在细胞生长与增殖、系统发生以及肿瘤发生等方面都有着重要的作用。CTCF的缺失会导致多种细胞分化和组织发育缺陷,包括影响记忆和学习能力、心血管发育、血液细胞分化以及肢端发育等。CTCF通过建立细胞谱系特异染色质高级结构调控决定细胞命运基因表达的分子机制,在多种生物学程序中发挥关键作用。
