BCAT2

支链氨基酸转氨酶2(BCAT2)是一种线粒体酶,主要负责亮氨酸、异亮氨酸和缬氨酸三种支链氨基酸的代谢,通过催化其转氨基反应生成相应的支链α-酮酸。该酶在肝脏、骨骼肌等能量代谢活跃的组织中高表达,因其线粒体定位特征,也被称为BCATm。研究发现,BCAT2通过调控支链氨基酸分解代谢,影响细胞能量供应和蛋白质合成,其异常表达与代谢综合征、糖尿病等疾病密切相关。近年来在肿瘤领域,BCAT2被发现可通过维持白血病干细胞内支链氨基酸水平,促进急性髓系白血病进展,成为潜在治疗靶点。目前针对BCAT2的药物开发尚处于早期阶段,有研究尝试通过小分子抑制剂阻断其酶活性,但尚无进入临床试验的候选药物。值得注意的是,由于支链氨基酸代谢与mTOR信号通路的交互作用,针对该酶的调控可能为代谢性疾病和癌症治疗提供新思路。
