Our default final concentration of glycerol is 50%. Customers could use it as reference.

The generation of the recombinant human melanoma-associated antigen 4 (MAGEA4) includes the fusion of the 10xHis-tag gene with all genes encoding the human MAGEA4, the cloning of the synthesized gene into the plasmid vector, the transfection of the cloned vector into the mammalian cells. This recombinant human MAGEA4 protein was quality tested in the purity, endotoxin level, and bioactivity. Its purity reached up to 90% measured by SDS-PAGE. The LAL method estimated the endotoxin content of this MAGEA4 protein less than 1.0 EU/ug. In the functional ELISA, this MAGEA4 protein can bind to the recombinant antibody against MAGEA4, with the EC50 of 30.33-43.10 ng/mL.
MAGEA4 is up-regulated in various malignant tumors such as oral squamous cell carcinoma, non-small cell lung cancer, pancreatic cancer, and breast cancer. It is not expressed in adult normal tissues but in germ cells. MAGEA4 plays a dual role as a tumor suppressor and tumor promoter during tumorigenesis. As a tumor promoter, MAGEA4 is highly expressed in liver cancer cells and inhibits caspase-mediated apoptosis, reduces the sensitivity to chemotherapy, and ultimately reduces p53 activity, thereby inhibiting apoptosis. MACEA4 acts as a master regulator of Tregs development and function. Tregs are involved in tumor escape from the immune system. MAGEA4 in pancreatic cancer inhibits T cell proliferation, thus facilitating tumor cell survival. As a tumor-specific antigen, MAGEA4 is helpful to establish a new antitumor method.
