I want to use Fish Igf-1 ELISA kit for fish's liver homogenate.
In the protocol of the kit, for tissue homogenates, after homogenizing, stored overnight at -20oC. After 2 freeze-thaw cycles, centrifuge the homogenates.I do not understand too much about this. It means I have wait for 2 days to homogenate 2 sample (homogenate-> store -20oC-> thaw-> store 20oC -> thaw-> centrifuge)? Can you explain for me. Thanks!

CSB-E12122Fh is no need to dilute generally.
So there is no sample diluent included in this kit.Pls let me know if you have any further questions. Thank you.
In the current protocol nothing is mentioned about an extraction of IGF-1 from binding proteins. This is generally a necessary procedure to properly measure IGF-1. Is an extraction, and if so which, recommended by Cusabio? Is this procedure in the most recent protocol?
Because this kit employs the competitive inhibition enzyme immunoassay technique and detects the total IGF1. Pls let me know if you have any further questions. Thank you.

功能:The insulin-like growth factors, isolated from plasma, are structurally and functionally related to insulin but have a much higher growth-promoting activity. Acts as a ligand for IGF1R. Binds to the alpha subunit of IGF1R, leading to the activation of the intrinsic tyrosine kinase activity which autophosphorylates tyrosine residues in the beta subunit thus initiatiating a cascade of down-stream signaling events leading to activation of the PI3K-AKT/PKB and the Ras-MAPK pathways. Binds to integrins. Its binding to integrins and subsequent ternary complex formation with integrins and IGFR1 are essential for IGF1 signaling.
